No Ashoka did not know about Luke as of now (Mandolorian season 2). But it is possible that in the Ashoka series we might see them interacting which would be awesome. A conversation between the apprentice and the son is something worth waiting for.

R2-D2 was present when Padmé gave birth to Leia and Luke, so he knew Luke was Anakin's son – but what he didn't know was that Anakin became Darth Vader. In addition to that, Anakin's fall was surely a painful event in R2-D2's life, one that he definitely wouldn't like to remember or even talk about.
She also became one of the few Jedi to survive a duel with the droid General Grievous. C3PO was clearly created from some sort of kit or existing prototype since every protocol droid aside from coloring looks the same. He didn't recognize him because again, they all look the same & it was decades until he saw him again. There was no reason to believe he or R2 even existed anymore.
He told the Death Watch warriors that Ahsoka was his betrothed, a ruse the Padawan reluctantly played along with. In Death Watch's camp, Ahsoka warned him that he was unwise to trust the Mandalorians. Mindful of their cover story, Lux quieted her by grabbing her and kissing her.
Overall, analyzing Star Wars Rebels shows that Jacen Syndulla was conceived in the days before Kanan's death. Still, Kanan's legacy lives on within Jacen. Even though Hera has been reluctant to have Jacen train as a Jedi, that reluctance might change with the return of Ezra Bridger to the main Star Wars galaxy.
