In this game, Peter Parker is now 25 years old with a degree in biophysics. He is living in Queens with Mary Jane after losing his aunt to a toxic gas caused by his own ex-mentor, Dr. Octavius. His girlfriend, Mary Jane, is living her dream as one of the best journalists in the city.
The "B." in Peter B. Parker stands for the character's usual middle name across his different versions, Benjamin. For Into the Spider-Verse producer and screenwriter Phil Lord, it also stood for "B grade or B-List or B-picture," to accentuate that Peter B. Parker was a lesser Peter than the one from Miles' universe.
Then, Miles further revealed that Across the Spider-Verse takes place one year and four months after the first one. Hence, he could be around late 14 or touching 15 in Spider-Verse 2. Since Gwen is 15 months older, she'd be 16 years or above in Spider-Man: Across the Spider-Verse, if not 17.
It's still plausible that Miles was 13 years old in Into the Spider-Verse and a freshman student at Visions if his birthday was between December and March. So if he's a sophomore, Miles was, at minimum, 15 years old in Across the Spider-Verse, which also meant, at minimum, Gwen would be 16 years and three months old.
Since Miles and Peni are the same age in the first movie, both being 13, it can be assumed that in the sequel she is the same age as Miles, who is now 15. However, due to placement of birthdays within a year, it is possible she is only 14 instead.

In Across the Spider-Verse, we learn that Peter B. made things right with his wife, Mary Jane, and the two had a child together. Peter B. expresses hope that his daughter will turn out to be like Miles.
Deadpool is in his late thirties/early forties, according to his co-creator Fabian Nicieza. Deadpool's age is difficult to determine due to his healing factor and Marvel's sliding timescale, but Nicieza narrowed down his age range, answering a question asked by many fans of the character.
