Originally incarnated as the Thirteenth Prime, Optimus died saving Prima from Liege Maximo. He was reincarnated into a innocent bot known as "Orion Pax". Optimus Prime and Megatron being brothers is heavily emphasized throughout the book. Ratchet even asks Optimus if he can "destroy his own brother." It's never stated how the AllSpark actually ended up on Earth in the first place. Optimus speaks to Sam outside his house in broad daylight.

Why is Megatron obsessed with Optimus Prime?
Megatron will never forgive Optimus for "betraying" him, stealing from the Council the rank of Prime; which was rightfully his. He has reserved for his old friend the greatest honor possible; glorious death at his hands. Megatron has been gone for three years; but now he has returned.
In G1, Megatron is equal to Optimus in a one on one fight. He beats Optimus a couple of times (once fatally). In Animated, Megatron was nearly scrapped by a bomb Starscream planted on him, and he was still almost too much for the Autobots. And shortly after he was rebuilt, he nearly wiped out the lot of them.
In this new Transformers Saga: Rodimus, Optimus Prime's son, wants to become a true Prime like his father. Optimus Prime and Rodimus eventually restore peace to the Universe, after defeating Megatron and the Decepticons once and for all!!!

His killing of Ironhide and the defeat of Optimus Prime establish how huge of a threat Sentinel Prime is. He even defeats Optimus twice, both times through fighting ability alone.
When the council approved Orion's ideas they renamed him Optimus Prime and told him he was now leader of all Cybertron. Megatron, furious that he was robbed of his rightful title believed Optimus had used him for a personal power play.
