Gamora Zen Whoberi Ben Titan was a former Zehoberei assassin and a former member of the Guardians of the Galaxy. She became the adopted daughter of Thanos and adopted sister of Nebula after Thanos killed half of her race. Gamora served him for years before betraying him in an attempt to free herself from his ways.
Thanos loaned the services of Gamora and Nebula to Ronan to aid him in the task. Realizing the mass casualties that would result should Ronan or Thanos gain control of the Orb, Gamora decided to betray them, accepting a high-priced offer from the Collector to retrieve the Orb for him.
Nebula realized that for her to truly live, Thanos would have to die. She wanted to get money to purchase weapons, so she attempted to steal Anulax Batteries from the planet Sovereign. However, the Sovereign people caught her and arrested her.
This was part of a cosmic process by which the wielder of the Soul Stone had to sacrifice something they truly loved. Thanos killed Gamora regretfully, showing he would truly do anything to accomplish his goals, no matter how much it hurt him personally.
The Variant Gamora was just pushed into a timeline she didn't comprehend and never really had the time to think about her complicated relationship with Thanos. Gamora survived Tony's snap because she had changed allegiances when it happened. The stones can interpret the users' intentions, and Tony only snaps Thanos' invading forces. As a youth, Nebula grew up in a torturous environment created by Thanos. Having the Mad Titan as a parental figure would have been bad enough, but she was also constantly compared to her surrogate sister Gamora. Thanos openly favored Gamora, which drove Nebula mad with jealous rage.
Thanos never cared much for Nebula, even going so far as to say in Guardians of the Galaxy that Gamora was his favorite. That could be because, to Thanos, Nebula was nothing more than a means to spare Gamora while still obtaining the Soul Stone.
