Because Bruno has stepped away from the family, the light on his door is dim, indicating that Casita's magic doesn't work in there anymore. Just like any other room, though, it was once enchanted and Bruno's room may serve as a metaphor for the heartbreak he endured trying to protect Mirabel.

Who can control Casita?
It has been theorized a lot of what Mirabel's gift is; a highly thought theory says her "gift" would be the same as Abuela Alma (being in charge of the magic). A part of this theory is that Mirabel and Alma both frequently control Casita in the movie (although it's likely that everyone can control Casita).
Thus, Abuela treats Mirabel harshly because she treats herself harshly. She puts an undue amount of pressure on herself to keep the family going in times of crisis by performing the role of a strong figurehead, trying to overcompensate with a sense of power and control because she had none over her husband's death.
Indeed, the reason why Mirabel doesn't get a gift in Encanto is that she already had the brains to take over as the family leader after Abuela Alma. Therefore, the miracle didn't need to give her an additional gift — Mirabel's gift in Encanto is her intellect, which she already possessed before the movie even began.
Bruno hadn't used his abilities for the last 10 years (it said he left when Maribel was 5 she's 15 now) so since he hadn't used it the magic in his room died out that would also be why the Casita couldn't help in there and why his room wasn't glowing.

Normally, Madrigals get their own rooms on the day they receive their magic gifts. But since Mirabel didn't get a gift, she didn't get a special room either.
She was unaware it was Bruno in the walls, she thought it was the rats talking in the walls. She also mentions in the his song that she associated him with the sound of sand, which his room constantly streamed with. Well her power is superhearing and she did says she heard him.
Encanto has a wealth of evidence that Casita is truly Abuelo Pedro. In addition to the house mirroring Abuel Pedro's sacrifice, the film lingers on Abuelo Pedro's portrait during the collapse, perhaps hinting that he's the person behind the magic.
