In the long history of the DC Universe, one of the most powerful beings to ever exist is, naturally, the daughter of Superman and Supergirl. But since the two heroes are Kryptonian cousins, their superpowered offspring has a forgotten Superman-family replacement to thank for her creation.
Did Superman and/or Supergirl ever get children? In the past, in certain Else Worlds comics, Superman has. Supergirl, as far as I'm aware, has never had any children in any form. In current comic line, Superman and Lois Lane do have a half-human and half-Kryptonian child name Jon.
On New 52 DC Multiverse Earth-38, Supergirl is Kara Kent, the daughter of Superman and Lois Lane on an alternate Earth where Superman and Batman began their careers in the 1930s, married in the 1940s and had children in the 1950s.

Supergirl season 1 set up Kara's romantic interest in James Olsen. The two were coworkers at CatCo and James had the benefit of already knowing her secret identity as Supergirl and it seemed like the pair were headed in the direction of potentially being an endgame relationship.
Portrayed by Elizabeth Tulloch, Lois is a reporter for the Daily Planet and the wife of Clark Kent / Superman. Tulloch also played alternate versions of Lois Lane in the Arrowverse television series The Flash, Supergirl, Arrow, Batwoman, and DC's Legends of Tomorrow.
In Peter David's Supergirl series, this Linda Danvers finds herself in another alternate reality with a Silver Age Superman. When he discovers that this Supergirl isn't really his cousin, they fall in love and get married. It's not weird. I mean it.
Kara and Trent continue to age, and after having sex with each other, Kara gets pregnant at a rapid pace. She gives birth, but the baby dies due to the time rapidly passing. The attempts to leave grow increasingly tense as Charles's worsening schizophrenia causes him to kill Brendan.
During the Invasion Mon-El and Kara had an affair which led to Kara's pregnancy with Mon-El's child. Mon-El kept the child a secret from his mother. Rose was born on Earth during the invasion of the Daxamites.
During the 1994 Zero Hour event, Power Girl experiences a mystical pregnancy and gives birth to a son (supposedly named Equinox), who ages rapidly. Finally he disappears, and has never been mentioned again in DCU.
Breeding between Kryptonian explorers and this race created a new Kryptonian hybrid race that could interbreed with a larger number of humanoid races—including Earth humans.
While multiple iterations of Superman Junior exist, his most notable iteration would probably be from “World's Finest Comics” #215 (1973). In here, Superman Junior is revealed to be the son of Superman with an unnamed human wife. Being a half-human, he possesses half of all the abilities of his father.
By all accounts Supergirl's marriage to Salkor was a happy one, and learning of his cousin's happiness with Salkor helped Superman come to terms with her death as well.

One surprise has been the revelation that Supergirl and Robin once went out on a date, which ended in disaster. Sadly, the date happened off-panel, but it raises so many questions.

Rose Zor-El also known as Rose Matthews is the daughter of Mon-El (Valor) and Kara Zor-El (Supergirl). She is the cousin of Clark Kent (Superman) and the granddaughter of Alura Van-El, Rhea and Lar Gand.
Despite being more youthful in appearance than her cousin, Superman, Supergirl is actually older than him. Though it hasn't always been the case, most versions of the two have interpreted Kara as being chronologically older than Clark, while being biologically younger.
