While it's never explicitly stated, Blade is seen with a large supply of Serum, in much smaller bottles from the first film. The amount of these he has suggests he does not need to increase his dose - Chances are, once she developed this, Karen left - either to return to a normal life or to join another group.

Does Karen turn into a vampire in Blade?
She becomes Blade's girlfriend, and join him in his quest to kill Deacon Frost. Unfortunately, Deacon Frost bites her, turning her into a vampire, so she leaves Blade so she can use her vast scientific knowledge in finding a cure for her condition.
Could be any number of reasons. I think the most logical is her character simply wasn't needed. She served her dual purpose in the first film of providing a cure and being an excuse to explain the world of Blade to the viewer.
