The answer is in the film. The story Barbossa tells Elizabeth. All pirates who took a piece of the treasure had to repay in blood, which is why Jack Sparrow cut his hand before tossing the medallion to Will in the cave.
Jack and Barbossa fought a fierce battle around the treasure cave until Jack stabbed Barbossa, who then pulled the sword out and stabbed Jack with it. However, once Jack stepped into the moonlight, he turned into a skeleton, revealing that he was cursed; having secretly palmed a piece of the Aztec gold.
Right before the film's climactic battle with the pirates at Isla de Muerta, Sparrow swipes a cursed coin from the treasure chest, making himself immortal and capable of dueling Barbossa. He shoots his nemesis with the pistol he has carried for ten years just as Will breaks the curse, killing Barbossa.
The crew of the Black Pearl needs Will Turner's blood to lift the curse because they killed his father. This would be unnecessary if Will's father never partook of the gold, and if he did, he would be subject to the curse himself, which is that he cannot die.
Elizabeth bids Jack, Barbossa, and the pirates farewell before reuniting with Will for their "one day" on the nearby island where their marriage is consummated and their only child, Henry Turner, is conceived.

Ultimately, the blood of Bootstrap's son was used to lift the curse. When Will reversed the curse by returning the last Aztec coins to the stone chest, he technically killed his father because at that moment, somewhere at the bottom of the ocean, Bootstrap Bill became mortal and would have drowned.
