In 2002, Marko and his accomplice Dennis Carradine robbed a wrestling venue while Flint, attempted to steal Uncle Ben's car to make his and Carradine's escape with the cash. Ben reasoned with Marko, but Carradine's arrival shocked him and he shot Uncle Ben at point blank accidentally, much to his horror.

Is Madame Web a hero or villain?
As envisioned in the comic books, Madame Web is not a villain. Instead, she's a hero who fittingly aids Spider-Man using her clairvoyant abilities. In the comics, Madame Web's real name is Cassandra Webb, and her psychic powers are due to her being a mutant like the X-Men.
Madame Web never truly fleshed out the morally ambiguous aspects of Ezekiel Sims, a villain who not only went head-to-head against Spider-Man, but also proved to be an unlikely ally and mentor to him.

And for the MCU's own Peter Parker, it was the untimely end of his beloved Aunt May, after she was caught in the blast of one of Green Goblin's pumpkin bombs, as seen in No Way Home. However, due to pandemic-era constraints, the scene in question was not shot as originally planned.
