Padmé's death was always a part of Palpatine's plan to turn Anakin to the Dark Side--but first, they had to be reunited. Being a master manipulator, Palpatine knew about Anakin and Padmé's secret relationship from the very beginning.
Because of Anakin's attachment to his wife, her potential death was the perfect excuse to step in and make false promises about the Sith. Palpatine even offered a way to save Padmé, and that the Sith could help preserve life. Of course, he never intended to save her, as she'd complicate his plans for Anakin.
It was also part of Darth Sidious' plot to ensure the passing of the Military Creation Act, of which Amidala was one of the leading opponents. The first attempt initially appeared to be a success, but Senator Amidala was found alive. The second attempt, however, was foiled by Jedi Obi-Wan Kenobi and Anakin Skywalker.
It is clear that he knows about it by the time of ROTS, where he exploits Anakin's concern about Padmé's potential death in childbirth to turn him to the dark side.
First, Palpatine did not know the truth about what had happened to Padme. But it was more fun to tell Vader that he had killed her to anger him. Padma was buried with her belly still appearing as if she was pregnant. To any Imperial observers, it would have appeared she had died before birthing any children.

Did Anakin regret killing Padme?
Anakin does not have even the slightest bit of remorse. By the end, he feels nothing but anger. Although he feels regret for killing Padme, he still helps the evil Emperor. The film ends with Anakins standing side by side with the man on whose behalf he has committed all his crimes.

It all begins with Palpatine, who wants to kill Padmé in order to drive Anakin closer to the dark side. He wisely tasks Count Dooku with taking her out because Palpatine doesn't want her death tied back to him.
Then, Vader also opened the Doors of the Dark Side and entered. Once inside the spiritual realm, Vader went through various trials, but eventually, he found Padmé. All indication was that Vader could have brought her back to life just like Momin did with his body. But Padmé refused to come.
He chose evil to achieve what he thought was the ultimate good. , this led to Padme's death. He choke the one he wanted to save with his own hands. When we turn a good thing into the ultimate thing and try to get it at all costs, we lose that good thing — destroy it with our own hands.
Anakin's Force Choke begins the series of injuries that ultimately leads to Padme's death, and convinces Obi-Wan to fight him. Despite this, when Anakin learns of Padme's death, his immediate reaction of grief and regret shatters everything in the lab with a powerful force shockwave.

Why did Anakin stay to serve Palpatine after Mustafar? Anakin really didn't have a choice. If he attempted to leave, and start a new life, Palpatine would just send the Imperial Military to kill him. If he killed Palpatine, Tarkin would probably send the entire Imperial Military to kill him.
Anakin's vision did come true, as Padmé, heartbroken by her husband's downfall, loses her will to live and succumbs while giving birth to twins. Before her death, Padmé still believed there was still good left in Anakin and that one day he could be saved with the help of their children.
In the main Star Wars timeline, of course, Darth Vader's attempts to resurrect Padmé on Mustafar were doomed to failure. But it's fascinating to speculate what would have happened had he been successful. In that scenario, Padmé Amidala's return from the dead would not have provided the solace Darth Vader sought.
This fell in line with various fan theories and non-canon novelizations that suggested Obi-Wan and Padmé had an affair sometime during the Clone Wars. In the Star Wars prequel movies, Obi-Wan often has a duty to protect Padmé, but there was never any sense of romantic feelings.
Every time she speaks, he remembers Padmé. Fighting alongside Sabé reminds him of fighting alongside his wife. While that is certainly painful enough, it's also reminds Vader of the man he used to be, a past he had long ago attempted to kill and bury. However, the memory of Padmé via Sabé won't let his past stay there.
Darth Vader did not 'forget' he was Anakin. He believed he was not Anakin Skywalker; a completely different person whom he killed. You will read in EU materials, that he referred to his memories as Anakin, as 'The Jedi'. He suppressed most of Anakin Skywalker's memories - his memories while he was Anakin.

Luke and Leia weren't Clones either, which is the other means of reproducing in SW. They were non-identical twins, they weren't grown in a vat like the Clone Troopers but born as a result of a normal pregnancy. So yeah, Anakin and Padme obviously had sex at least once, but since SW is for kids we don't need to see it.
