R2 knows everything. Theres a weird misconception that R2 had his memory wiped as well, he didnt. He knows everything and everyone that was a part of The Skywalker Story. He just never said anything to protect luke and leia.
R2-D2 has not had a memory wipe since at least the start of the Clone Wars, and was present for most of the main events in the first 6 movies. We can assume he knew pretty much everything there was to know, including that Anakin married Padme, that Leia and Luke were Anakin's kids, and probably that Anakin was Vader.
Darth Vader may have never known Princess Leia was his daughter in Star Wars and never cultivated a relationship with her, even when he was a Force ghost. Anakin Skywalker never knew Padmé was pregnant with twins, and he believed his child had died with his wife.

Darth Vader acts like he doesn't even know C-3PO or R2D2. They were with him since he was a child. He did everything he could to forget his past. Also, his plating had been replaced many times, and protocol droids all look very similar.

Did R2-D2 know that Luke was Anakin's son?
R2-D2 was present when Padmé gave birth to Leia and Luke, so he knew Luke was Anakin's son – but what he didn't know was that Anakin became Darth Vader. In addition to that, Anakin's fall was surely a painful event in R2-D2's life, one that he definitely wouldn't like to remember or even talk about.
The sequence calls back to a similar scene in Revenge of the Sith, when Bail Organa orders Threepio's memory be erased to prevent him from sharing information about Anakin Skywalker's fall to the dark side.
Maybe R2-D2 didn't know Darth Vader was Luke's father because he didn't see Palpatine christening him Vader, or maybe he believed Anakin was dead because even Obi-Wan didn't know Anakin was alive.
C3PO was clearly created from some sort of kit or existing prototype since every protocol droid aside from coloring looks the same. He didn't recognize him because again, they all look the same & it was decades until he saw him again. There was no reason to believe he or R2 even existed anymore.
Leia's Force abilities and her lack of awareness of her true identity also contributed to Vader's inability to sense their familial connection. Vader's denial of Anakin Skywalker's legacy and his fear of getting hurt again prevented him from realizing the truth about his children until later in the original trilogy.
Because Bail Organa ordered C3PO's memory wiped at the end of the Revenge of the Sith. This was to protect the twins identity because he knew that C3PO had a tendency to run off at the mouth!
secondly, in Legends, R2 was given programming that actually restricted him. from sharing any information with Luke or Leia. about their parents or relation to one another. this was all part of the plan. to conceal this information from the twins.
in the Dark Horse comic series star Wars Tales, which is now part of legends, it's revealed that Darth Vader did recognize C. 3PO on Cloud City. when 3PO was shot by Stormtroopers, his parts were brought before Vader. because the Imperials thought he could hold valuable.
The question being answered is: “Do R2D2 and C3PO know that Darth Vader is Anakin?” C-3PO? Definitely not. As many other have pointed out, C-3PO had his memory wiped at the end of Episode III, so it is almost certain that he does not know.

The name derives from when Lucas was making one of his earlier films, American Graffiti. Sound editor Walter Murch states that he is responsible for the utterance which sparked the name for the droid. Murch asked for Reel 2, Dialog Track 2, in the abbreviated form "R-2-D-2".
