Gothel's burning desire to retain her youth for all eternity has notably driven the woman to complete madness, to the point where she was more than willing to force Rapunzel into slavery for the rest of her years when she figured out the truth of their relationship.

Did Mother Gothel stab Flynn?
After Mother Gothel stabbed Flynn Rider (who was actually named Eugene Fitzherbert), Rapunzel made a deal with her mother to save his life. She promised that she would go with Gothel anywhere she wanted and wouldn't try to escape, so long as Rapunzel was allowed to heal his wounds.
Although she did take care of her when growing up and did gain her affection through treats, birthday gifts and pretense of motherly love, she spent eighteen years subtly but viciously abusing Rapunzel, verbally and emotionally, to destroy her self-esteem and make her completely dependent and vulnerable on her.
According to Flynn Rider, she has been using it for hundreds of years. When the queen uses it, Mother Gothel wants to get it back, so she breaks into the palace and cuts off a lock of Rapunzel's hair to keep for herself.
However, Mother Gothel betrays them and knocks the duo out with a branch, making it seem as if she was rescuing Rapunzel as her entire plan was to keep Rapunzel with her, hidden away in their tower in the forest. Offscreen, the brothers are arrested and taken to the prison along with Flynn.
