Jake Sully, ex-Marine, is recruited into the RDA's Avatar program after his scientist twin-brother, Tommy, is killed. Since Jake and Tommy share identical DNA make-up, Jake is able to step into his deceased brother's role as an Avatar driver.
In the theatrical release of the film and the extended re-release, Tom was said to have been killed by a gunshot while in the Extended Collector's Edition, Jake said that Tom's killer used a knife. The name Tom means "Twin" and Tom was Jake's twin brother.

Why was Neteyam killed off?
It was only when Lo'ak insisted that Neteyam agreed to the rescue. Neteyam risked his life and perished in order to save Lo'ak and Spider from Quaritch. As Jake himself said: "A son for a son". Spider wept for Neteyam as he died as well.
