In 0 BBY, Lars and Luke Skywalker purchased the astromech droid R2-D2, who, unbeknownst to them, possessed stolen plans for the Death Star, an Imperial superweapon. Imperial stormtroopers tracked the droid to Lars' homestead, where they interrogated and murdered both him and his wife.
It was the last day of Owen's life. Stormtroopers searching for C-3PO and R2-D2 tracked the droids to the Jawa scavengers who'd found them and learned to whom they'd been sold. They arrived at the Lars farm and interrogated Owen and Beru, killing them and leaving the homestead in flames.
Owen felt that Obi-Wan was personally responsible for the deaths of Shmi and Anakin and, that the Jedi Master's actions had ripped his family apart. Despite his dislike of the Jedi and Kenobi in particular, Owen did have honor, refusing to turn Kenobi in when several Sith Inquisitors came looking for him.
Owen Lars, however, was kept out of the equation and held strong resentment toward Obi-Wan, who told him that Anakin died during the night of Order 66, only to hide Vader's identity from the Sith Lord's stepbrother.
The stormtroopers who'd tracked the droids to the Lars farm murdered Owen and Beru when they realised that the droids were gone and the plans weren't anywhere on the property. Vader did not directly order their deaths - it would have been the officer, or more likely the squad leader, in charge of the unit.
