The Inquisitorius, also known as the Inquisitorius Program, the Order of Inquisitors, the Inquisitor corps, and the Imperial Inquisition, was an organization of Force-sensitive operatives under the Galactic Empire.
The hierarchy of the squad was quite simple: Vader oversaw the organization at large while the Grand Inquisitor was responsible for leading them in their day-to-day operations and reported directly to Vader. The rest of the Inquisitors reported to the Grand Inquisitor, but a constant struggle for power was apparent.
The first Inquisitor we meet is the head guy himself, The Grand Inquisitor. He appears in the opening scene of the first episode of Star Wars Rebels, taking orders from Darth Vader himself. We know more about him than most of his brethren. He was a Jedi Temple Guard before the Great Purge, a Pa'un by birth.
Inquisitors inducted into the ranks of the Inquisitorius left behind their old names and took up new ones, in which they were labelled Brother/Sister, denoting their sexes.

1 The Grand Inquisitor Was The First Shown And The Greatest
As the leader of the entire Inquisitorius, he was the most powerful and intelligent of the Inquisitors. He was brought to life in animation and live-action. While his live-action adaptation disappointed fans, his animated representation was chilling.
The first grand inquisitor in Spain was the Dominican Tomás de Torquemada; his name became synonymous with the brutality and fanaticism associated with the Inquisition.

Vader was apparently made Supreme Commander by a direct appointment from Emperor Palpatine, as was his son when Palpatine was reborn in a clone body. Xecr Nist and probably Sedriss before him were likewise appointed as Executors by Palpatine.
He then dueled Kanan and Ezra, who'd arrived with the other rebels to save their friend. This time, Kanan defeated the Inquisitor. Disarmed and humiliated by his failure, the Imperial agent plunged to his death – but warned Kanan he had unleashed something terrible by defeating him.
It became the most substantive of the three different manifestations of the wider Catholic Inquisition, along with the Roman Inquisition and the Portuguese Inquisition.

The most (in)famous Grand Inquisitor, Tomás de Torquemada, was, not surprisingly, the queen's confessor. After the Reconquista (the conquest of the Muslim regions in Spain), the newly emerging Spanish national state had no place for anyone other than the Christians.

As Palpatine's Sith apprentice, Vader was in charge of the Inquisitors, who quickly came to fear his power. While Vader was far more powerful than the Inquisitors, he was not nearly as sadistic as they were, as shown by their hunting methods and what they did with the Jedi they captured.
Why are there no Inquisitors in the original trilogy?
Recent Star Wars projects have heavily featured the Inquisitorius. But they are nowhere to be seen by the time of the original trilogy. An easy explanation is that many of the Inquisitors fell in combat. Marrok was revived and fought well after the demise of the Empire.
Once they had been corrupted to the dark side, the Inquisitors no longer went by their original names and instead were numbered, referred to only as Brother or Sister. While fans have seen at least 14 different Inquisitors across several Star Wars media, only ten have been officially named as a Brother or Sister.
