The spider, brought to Miles's Earth-1610 by the Into the Spider-Verse antagonist Kingpin's super-collider machine, was actually from Earth-42, leaving that world without a spider to bite Peter Parker, Miles Morales, or anyone else who could carry the mantle, and thus created a world without Spider-Man.
Notable differences. There is no Spider-Man (since it was supposed to be Miles originally). The overall color scheme of the dimension is different and consists of more red, green, and purple. All of New York is plagued with crime as there is no one to stop them.
Forty-two was Jackie Robinson's jersey number for the Brooklyn Dodgers. He was a barrier-smashing black superhero in baseball, and [Miles is a] color-smashing black superhero in Spider-Man comics.
Was the spider that bit Miles supposed to bite Miguel?
No. This is a different universe, so the rules have changed. In Earth 42, we see Miguel say that the radioactive spider that bit Miles was supposed to bite someone else, and since it didn't bite anyone from that universe, it was without a Spider-Man.

The main conflict of Spider-Man: Across the Spider-Verse centers around Miles Morales' refusal to experience the same fate of every other Spider-Man, which leads Miguel O'Hara a.k.a. Spider-Man 2099 to chase down Miles in order to keep the multiverse safe from collapse.

That spider—seen glitching prior to biting Miles in Into the Spider-Verse—wasn't from his world. That's why Miguel O'Hara calls Miles “the” anomaly.
The number 42 is, in The Hitchhiker's Guide to the Galaxy by Douglas Adams, the "Answer to the Ultimate Question of Life, the Universe, and Everything", calculated by an enormous supercomputer named Deep Thought over a period of 7.5 million years. Unfortunately, no one knows what the question is.
Miles Morales had never been bitten by a genetically enhanced spider and became Spider-Man? What if instead…the U.S. government recruited, trained, and granted him incredible powers with the Super-Soldier Serum? What makes Miles a hero no matter the circumstances, no matter the reality?!
He is the second evil version of Miles Morales, the first being his Earth-616 counterpart, better known as Ultimatum. According to the official Spider-Man: Across the Spider-Verse artbook, Miles G. Morales isn't technically a villain but more so a vigilante.
At some point prior to the Miles Morales of Earth-1610B accidentally arriving in Earth-42, Jefferson Morales had died. He had a large mural dedicated to him spraypainted by his son, Miles.
In 2019, after a battle with the Spider Society, the Miles who became Spider-Man was accidentally transported to Earth-42 after trying to return to his home dimension. Once he realized he wasn't home, he pretended to be the Earth-42 Miles to try and blend in, but Aaron and the Prowler saw through it and captured him.
If this hadn't happened, according to Miguel, the Peter Parker of Miles' world would have survived and the spider would have bitten someone else on Earth-42 (its home dimension) to give that world a Spider-Man. Instead, Miles became a Spider-Man who was never supposed to exist.
Finally, the other Spider-People frequently refers to Miguel as a vampire, mistaking his fangs for those of a bloodsucker. We can even see Miguel's fangs in use for a moment when Spider-Man 2099 bites the neck of the Renaissance Vulture to paralyze the villain and bring him to the Spider Society base.
In the beginning, when Spot's out of control, he ends up in his own little pocket dimension without any spots. Then, when he recreates a Collider and starts traveling from dimension to dimension, he becomes all black when the spots overtake his body, transforming into an abyss-like character.
The Spot, on the other hand, is incredibly petty and upset about being made fun of and viewed unfairly, to the point where this fuels his desire to destroy the universes of every Spider-Person ever, and even believes Miles' role in destroying the collider is what led to his new form.
But as it turns out, Miguel's “hatred” for Miles runs much deeper than Miles being a bit of a rule-breaker. Miguel explains that Miles is an anomaly, which makes it that much harder for Miguel to protect the fabric of the multiverse.
Hailey Cooper is a recurring original supporting character in Marvel's Spider-Man series, first appeared in Marvel's Spider-Man: Miles Morales. As of Marvel's Spider-Man 2, she becomes Miles Morales' love interest.
The movie reveals Miles' dad, Jefferson, has to die for Miles' journey to continue, as fate has dictated on his Earth-1610. However, Miles rebels against Oscar Isaac's Miguel O'Hara (aka Spider-Man 2099) and the legion of Spider-People in charge of protecting the multiverse.
According to Miguel O'Hara, there is only meant to be one Spider-Man per universe, and the spider biting Miles prevented another universe from having a Spider-Man and put Miles in a position where Peter Parker had to save him, thus costing him his life. Because of all of this, Miles' Spider-Man is an anomaly.
Prodigy-level geneticist Miguel O'Hara gives himself spider-like abilities in an experiment gone wrong. He sees an opportunity to protect ordinary people against abusive corporations, and decides to use his powers for good and become Spider-Man of the year 2099.
Miles' eagerness to be a hero is both his strength and weakness; it propels his desire to grow and assist others but also makes him prone to danger and reluctant to accept help. Miles also dislikes being a victim or receiving pity.
