Had Sam not confronted and injured the enormous spider, then she would've eaten Frodo. As well as having his life saved, Frodo was reliant on Sam's willpower and encouragement to keep them going. Just before they entered Mount Doom, Frodo collapsed and seemingly wasn't going to be able to make it to the fires.
Frodo literally would have died, likely along with Merry and Pippin (but not Sam, because he's heroic) before they even reached Bree. When scampering down the road, trying to escape the riders, the four hobbits hide under an overhang on the side of the road.
What would have happened if Sam didn't go with Frodo?

Sam would later discover that Frodo was merely paralyzed by the giant spider's venom and he was not taken by some orcs. Sam came to the rescue again and fought through the orcs to reach Frodo.

Frodo is the person who carries the Ring because of reasons which, ultimately, make him a completely ordinary person. Frodo's Hobbit nature allows him to be more incorruptible, discreet and all but invisible to Sauron.

Does Frodo send Sam away in the book? No, and this is another meaningless deviation from the source. This was presumably done for tension, but the book has enough of this.
Could Frodo and Sam have gotten through the black gate?
Until Gollum told them that there was another way into Mordor, Frodo and Sam were going to enter Mordor through the Black Gate. If they proceeded, they almost certainly would've been caught and killed.
He journeyed all the way to Mount Doom to destroy the Ring, but when he got there, he couldn't do it. That was because of the location. Sauron forged the One Ring inside of Mount Doom, which meant that its power was significantly most potent inside the volcano.
Though someone could start out with good intentions, the Ring would eventually corrupt them. And that is why Gandalf can't touch it. He is afraid that if he did, it would corrupt him and make him just as bad as Sauron since Sauron put so much of himself and his evil into the One Ring.
The One Ring had the power in The Lord of the Rings to corrupt the beings of Middle-earth just from being in close proximity. As a powerful wizard, Gandalf wasn't affected in quite the same way.
The One Ring has the power to extend the life of any mortal indefinitely. Gollum survived after losing the Ring because its effects on him lingered as long as the Ring existed in the world.
Because Sauron underestimated the will of the dwarves and the intelligence of the elves. The dwarves of Middle-Earth were notoriously strong willed, able to resist corruption and stay true to their purpose. Sauron had no means by which to corrupt them regardless of how he tried.
There's a big reason why this coincidence didn't clock with Gandalf, and it's because the idea of Bilbo finding the One Ring seemed out of the realm of possibility. For close to 3000 years, the One Ring was missing, with no hints as to its whereabouts.
As for Sam, his resistance likely has to do with his innate goodness and loyalty. His love for Frodo helped him to overcome the pull of the Ring and his inner pureness was never fully pierced by the Ring.
After his wife died in Fo. A. 61, on 22 September Sam left Bag End, and went to the Tower Hills where he was last seen by Elanor, entrusting to her the Red Book; according to her, he went to the Grey Havens to sail across the Sea and be reunited with Frodo in the Undying Lands.
Sméagol was of a race that is slightly different to hobbits themselves, which would explain both his susceptibility to the Ring's corruption and also his subsequent invisibility when putting the Ring on.
