A smart, spunky droid who would serve a multitude of masters over his lifetime, R2-D2 was never given a full memory wipe nor did he ever receive new programming, with these factors resulting in an adventurous and independent attitude.

A reliable and versatile astromech droid, R2-D2 has served Padmé Amidala, Anakin Skywalker, and Luke Skywalker in turn, showing great bravery in rescuing his masters and their friends from many perils.
Most are quite intelligent, and some can even speak the human language, such as C-3PO. However, most droids, including R2-D2 and BB-8, "speak" a droid language through sequences of beeps and whistles. It is known as "Binary," and several humans in the Star Wars universe can understand it.
By wiping his memory, it allowed the leeway necessary to bridge the logic gap created by the previous two prequel films. However, the same could not be said for R2-D2, who never had his memories erased.

Darth Vader acts like he doesn't even know C-3PO or R2D2. They were with him since he was a child. He did everything he could to forget his past. Also, his plating had been replaced many times, and protocol droids all look very similar.
Anakin Skywalker ignored this procedure in regards to R2-D2, to the chagrin of both the Jedi Council and the Republic Military leadership. The Galactic Empire did not grant its droids any independence and sought to keep them subservient and docile.
But only a few of the human characters in Star Wars can understand droids; Luke Skywalker wasn't the only one. Obi-Wan Kenobi was another human who could speak Binary, as well as Anakin, who shared the strongest relationship with R2-D2, as seen in the prequel trilogy and Star Wars: The Clone Wars.
Binary, also known as droidspeak or astromech, was the language commonly spoken by droids. It consisted of a set of beeps and whistles strung together into what resembled sentences.
After the Clone Wars, however, Brooks Propulsion Devices, the company responsible for the development of his boosters, was shut down, and his rocket boosters were eventually damaged and thus could not be repaired. Despite this, however, R2 hardly regretted the loss of his ability to fly.

Towards the very start of George Lucas' creation of R2-D2, both he and C-3PO had speaking roles, leaving R2-D2's trademark bleeps without a place in the story. This plan was abandoned eventually, but C-3PO does often explain to the audience that R2-D2's bleeps often translate to swear words.

in the Dark Horse comic series star Wars Tales, which is now part of legends, it's revealed that Darth Vader did recognize C. 3PO on Cloud City. when 3PO was shot by Stormtroopers, his parts were brought before Vader. because the Imperials thought he could hold valuable.
The question being answered is: “Do R2D2 and C3PO know that Darth Vader is Anakin?” C-3PO? Definitely not. As many other have pointed out, C-3PO had his memory wiped at the end of Episode III, so it is almost certain that he does not know.
